When you say MOS is dependent on the stealth "spec" do you mean the level of stealth in which actual points were spent or the composite stealth skill including +stealth from items and realm rank?

That is, if I spec 34 into stealth via training points, but have +5 stealth from items and +1 from realm rank would my Mastery of Stealth level be 34 for level 6 MOS (+25% speed) or 40 for MOS 7 (+28% speed)?

Do we know this for a fact? Also, is there any difference besides the movement speed bonus between the different MOS levels? That is, can you see someone at a greater distance if they have a lower overall MOS level?

Judging by the wording here, I would assume that a combined 50 stealth would grant you max MoS

Assuming 50 comb spec stealth for everyone:
Assassins see other assassins at a range of 125.
Assassins see archers at a range of 250.
Assassins see minstrels at a range of 250.
Archers see assassins at a range of 125.
Archers see other archers at a range of 125.
Archers see minstrels at a range of 125.
Minstrels see assassins at a range of 125.
Minstrels see archers at a range of 125.
Minstrels see other minstrels at a range of 125.

Base spec is what determines the MoS level. MoS currently does not provide any range increase, only movement speed. The listed visibility ranges are as they always were with detect hidden without see hidden / mos / any other view distance increasing stuff.
